Accelerated diabetic wound healing by topical application of combination oral antidiabetic agents-loaded nanofibrous scaffolds: An in vitro and in vivo evaluation study

The combination of oral antidiabetic drugs, pioglitazone, metformin, and glibenclamide, which also exhibit the strongest anti-inflammatory action among were loaded into chitosan/gelatin/polycaprolactone (PCL) by electrospinning polyvinyl pyrrolidone (PVP)/PCL composite nanofibrous scaffolds pressurized gyration to compare diabetic wound healing effect. therapies significantly accelerated in type-1 rats organized densely packed collagen fibers dermis, it showed better regeneration dermis epidermis than single drug-loaded with less inflammatory cell infiltration edema. formation hair follicles started 14 days only therapy lower proinflammatory cytokine levels observed compared treatment groups. increased wettability hydrophilicity scaffolds, demonstrated sustained drug release over days, has high tensile strength suitable cytocompatibility on L929 (mouse fibroblast) created a area for proliferation fibroblast cells. Consequently, application metformin pioglitazone-loaded chitosan/gelatin/PCL offer bioavailability, fewer systemic side effects, reduced frequency dosage amount drug.
